Article Abstract:
New Bacillus thuringiensis strains having extended insecticidal activity were constructed by transduction and transformation. The transduction technique used phage CP-54Ber to transfer recombinant plasmids containing cryIA(a) delta-endotoxin gene, resulting in 5 x 10(super -8) to 2 x 10(super -6) transductants per colony forming unit. In Bacillus thuringiensis subspeciesunable to accept transduced plasmids, electrotransformation was successfully used. The results showed that transduction and transformation can be used to extend the host specificity of Bacillus thuringiensis strains.

Article Abstract:
The nprA gene which encodes the neutral protease A of Bacillus thuringiensis was cloned through the application of gene-specific oligonucleotides. The size of neutral protease A determined from the nprA sequence was 566 amino acids. The cloned nprA gene was partially deleted in vitro, and the deleted allele, called nprA3, was employed to construct an nprA3 strain of B. thuringiensis. The nprA3 strain produced insecticidal crystal proteins that were more stable than those of the isogenic nprA+ strain.

Article Abstract:
The screening of Bacillus thuringiensis serotypes for the presence of cryV-type insecticidal genes and the cloning and sequencing of a cryV gene from B. thuringiensis subsp. kurstaki DSIR732 were reported. The nucleotide sequence revealed an open reading frame coding for a protein of 719 amino acids. The gene was expressed in Escherichia coli and the 81-kDa cryV gene product showed toxicity to Epiphyas postvittana (Lepidoptera: Tortricidae).
