Flow cytometric analysis of the in situ accessibility of Escherichia coli 16S rRNA for fluorescently labeled oligonucleotide probes

Article Abstract:

A study was conducted to carry out a flow cytometric quantification of fluorescent signals conferred by carboxyfluorescein-labeled oligonucleotides targeted to the 16S rRNA of Escherichia coli. The oligonucleotide probes complemented the sequence determined from E. coli and were synthesized with 5-(6)-carboxyfluorescein in the last step of solid-phase synthesis. Results indicated differences in the primary and higher-order structures. Findings also showed that some probe target sites were open and can be deleted.

Potential interactions of particle-associated anammox bacteria with bacterial and archaeal partners in the Namibian upwelling system

Article Abstract:

Comparative 16SrRNA gene analysis and catalyzed reporter deposition fluorescence in situ hybridization (CARD-FISH) was used for the identification and particle association of anammox bacteria in Namibian oxygen minimum zones (OMZ). Findings revealed that the typical marine anammox bacteria, Candidatus Scalindua, colonized microscopic particles which were the remains of either microscopic marine snow particles or resuspended particles.

Article Abstract:

Research has been conducted on Escherichia coli 16S rRNA. The authors have compared the in situ accessibility of this rRNA for Cy3-labeled oligonucleotide probes with the 30S ribosomal subunit's 3D-structure model, and describe the effect of fixation methods and hybridization procedures on 16S rRNA in situ accessibility fo E. coli.
