A RACHITT for our toolbox

Article Abstract:

The authors used RACHITT (random chimeragenesis on transient templates) to create a hybrid dibenzothiophene monooxygenase that is more efficient at biodesulfurization of fossil fuels than parent genes to show the effectiveness of this method of gene shuffling. The article also refers to "DNA shuffling method of regenerating highly recombined genes and evolved enzymes" of this issue (p. 354-359).

Sequence-activity relationships guide directed evolution

Article Abstract:

A combined computational and experimental method, which incorporates a protein sequence-activity relationship (ProSAR), is used for evolving proteins with novel properties. The ProSAR approach is used to analyze the effects of multiple mutations and also identify beneficial mutations.
